The coding region has been updated to extend the N-terminus to one that is more supported by the available transcript and conservation data. It should be noted that an alternative downstream start exists, which would result in a protein that is 6 aa shorter at the N-terminus. Both start codons are well-conserved and have weak Kozak signals, so it is possible that leaky scanning by ribosomes will allow the use of the downstream start codon at least some of the time. SignalP3.0 predicts the presence of an N-terminal signal peptide in both the longer and shorter proteins, though the probability is slightly higher for the shorter shorter. There is currently no experimental evidence indicating which start codon is preferentially used in vivo. |
