The coding region has been updated to extend the C-terminus following the addition of several exons at the 3' end. There are no full-length transcripts representing the update, but partial transcripts do span its length in both human and the mouse ortholog. The updated sequence includes a large exon 10 that has no single transcript spanning its full extent. Nonetheless, BX648206.1 covers most of this exon, as does AB037748.2, which is spliced to the 3' neighboring exons. AF528529.1 and AK090534.1 are spliced to the 5' neighboring exons, supported by orangutan CR860680.1. Several tiled transcripts also span this exon in mouse, including AK140021.1, AF013969.1 and AK122495.1. Some transcripts appear to have polyA tails and terminate in this large exon, e.g., BX648206.1, AK025965.1 and AF528529.1, but on closer inspection, it appears that these may be priming off internal 'A' nucleotides in exon 10, and they do not have polyA signals. The open reading frame represented in the update also shows good conservation in models from other species, including macaque XP_001098928.1, dog XP_536235.2, cow XP_585315.3, chicken XP_420784.2, and rat XP_001060944.1. It is also of note that the genomic alignments between these species show insertions/deletions in the region of exon 10 that are always in multiples of three, or else they are compensated in order to restore the reading frame. This is a strong signature of a protein-coding region and provides good supporting evidence for the validity of this exon/intron structure. |
