Background: The Limulus polyphemus DNA used for sequencing was derived from a single male animal (carapace length approximately 16 cm) collected January 2008 in Woods Hole, MA. Muscle tissue was extracted from the limbs, and genomic DNA was prepared ... in the lab of Nipam Patel by Meredith Protas. Total sequence coverage of Roche 454 reads was approximately 18C (fragments, 3 and 8kbs) using a genome size estimate of 1.5Gb. The combined sequence reads were assembled using the Newbler software (Roche). This draft assembly was referred to as Limulus polyphemus 2.1.1. This version has been gap filled, error corrected with 12X Illumina reads, and cleaned of contaminating contigs. The assembly is made up of a total of 286,840 scaffolds with an N50 scaffold length of 238kb (N50 contig length was 11.4kb). The assembly spans 1.7Gb.
