The objective of this study was to identify the genetic markers in einkorn (monococcum) wheat RIL population and use those markers to construct the genetic linkage map. Here, we performed skim-sequencing of the RIL population using Nextera library and whole genome sequencing of the RIL parents accessions using TruSeq method. We demultiplexed the sequencing data for both libraries and trimmed the fastqs to remove adapters and any unwanted sequence parts. We called the genotypes of the population and parents using two einkorn assemblies. Then, the genetic linkage map was constructed by generating the recombination bins.
