Mouse embryonic stem cells expressing an Enhanced Yellow Fluorescent Protein (EYFP) transgene from one ROSA26 locus and LacZ from the other ROSA26 locus, as well as tamoxifen-inducible CRE recombinase from untargeted viral integration, were maintained on a mouse embryonic fibroblast feeder layer by daily replacement of mES media [KO-DMEM (Invitrogen, San Diego, CA) containing 15% knock-out serum (Sigma, St. Louis, MO), 1X glutaMAX (Gibco, San Diego, CA), 1 X non-essential amino acids (Gibco), 55 μm 2-mercaptoethanol (Gibco), and 1000 U/mL mLIF (Millipore, Billerica, MA)]. These cells were a gift from A. McMahon (Harvard) (Hayashi and McMahon 2002; Mao et al. 2005).
