DNA flanking Mu transposon insertions were amplified from transposon inactive, UniformMu seed mutants using thermal assymmetric interlaced PCR (TAIL). TAIL-PCR was completed with primers specific for the Mu terminal inverted repeat and a set of 12 arbitrary primers. Amplicons were size enriched using Sepharose 400 spin columns prior to cloning.
