The 3'anchored cDNA Sequences were generated using the 454-GS20 platform. cDNA was prepared from total RNA using a biotinylated dT-B adaptor to prime first strand synthesis. Double stranded cDNA was purified on streptavidin beads and digested with MspI. A custom A-adaptor that incorporated a 3 base multiplex key a CG overhang was ligated to the MspI ends. Following ligation, single stranded sequencing template was eluted and sequenced on the GS-20 instrument. Libraries were constructed from wild type and viviparous1 mutant ovaries and sequenced in a single multiplex reaction. Consensus sequences for anchored cDNA tags were obtained by CAP3 assembly.
