Description

The p.H343Y variant (also known as c.1027C>T), located in coding exon 2 of the RBM20 gene, results from a C to T substitution at nucleotide position 1027. The histidine at codon 343 is replaced by tyrosine, an amino acid with similar properties. This alteration has been reported in a hypertrophic cardiomyopathy (HCM) cohort; however, clinical details were limited (Lopes LR et al. Heart, 2015 Feb;101:294-301). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
