Regulatory logic of endogenous RNAi in silencing de novo genomic conflicts

(Submitter supplied) Although the biological utilities of endogenous RNAi (endo-RNAi) have been largely elusive, recent studies reveal its critical role in the non-model fruitfly Drosophila simulans to suppress selfish genes, whose unchecked activities can severely impair spermatogenesis. In particular, hairpin RNA (hpRNA) loci generate endo-siRNAs that suppress evolutionary novel, X-linked, meiotic drive loci. The consequences of deleting even a single hpRNA (Nmy) in males are profound, as such individuals are nearly incapable of siring male progeny. more...

Rapid evolutionary dynamics of an expanding family of Drosophila meiotic drive factors and their hpRNA suppressors

(Submitter supplied) The Dox meiotic drive system distorts the sex-ratio in Drosophila simulans. Here, the authors reconstruct the step-wise emergence, and recent amplification of Dox superfamily genes in parallel with the emergence of autosomal hairpin RNA-class siRNA loci that target subsets of these putative drivers

Simple and Complex Centromeric Satellites in Drosophila Sibling Species

(Submitter supplied) Centromeres are the chromosomal sites of assembly for kinetochores, the protein complexes that attach to spindle fibers and mediate separation of chromosomes to daughter cells in mitosis and meiosis. In most multicellular organisms, centromeres comprise a single specific family of tandem repeats, often 100-400 bp in length, found on every chromosome, typically in one location within heterochromatin. more...

Developmental hotspots drive transcriptional variability and convergence in the Drosophila olfactory system

(Submitter supplied) The goal of this study was to identify the preponderance of developmental hotspots in the Drosophila olfactory system. Using RNASeq-derived transcriptome profiles of the developing and adult antennae for six different Drosophila species, we show that a few highly variable transcription factors may drive, likely in a combinatorial fashion, high variability in a few select olfactory receptor neuron (ORN) developmental lineages (i.e. more...

A somatic piRNA pathway in the Drosophila fat body ensures metabolic homeostasis and normal lifespan

(Submitter supplied) In gonadal tissues, the Piwi-interacting (piRNA) pathway preserves genomic integrity by employing 23-29nt small RNAs complexed with argonaute proteins to suppress parasitic mobile sequences of DNA called transposable elements (TEs). While recent evidence suggests that the piRNA pathway may be present in select somatic cells outside the gonads, the role of a non-gonadal somatic piRNA pathway is not well characterized. more...
