MicroRNA array studies in Hirschsprung disease

Aging aggravates aortic aneurysm and dissection via the miR-1204-MYLK signaling axis

(Submitter supplied) The mechanism by which aging induces aortic aneurysm and dissection (AAD) remains unclear. A total of 430 subjects were recruited for screening of differentially expressed plasma microRNAs. We found that miR-1204 was significantly increased in both plasma and aorta of elder patients with AAD, and was positively correlated with age. Cell senescence induced the expression of miR-1204 through p53 interaction with plasmacytoma variant translocation 1, and miR-1204 induced vascular smooth muscle cell (VSMC) senescence to form a positive feedback loop. more...

Single-Cell Transcriptomics of Epstein-Barr Virus and Human Herpesvirus 6 Coinfection

(Submitter supplied) Epstein-Barr virus (EBV) and human herpesvirus 6 (HHV-6) infection is widespread among people. Here I describe single-cell RNA sequencing of two lymphoblastoid cell lines harboring both episomal EBV and inherited chromosomally integrated HHV-6. Rare instances of HHV-6 expression appear enriched with EBV reactivation.

Expression analysis of microRNAs in INT-HA-induced M2-like macrophages and untreated macrophages (M0)

(Submitter supplied) Using the highly sensitive miRNA microarray, we screened 107 up-regulated microRNAs and 33 down-regulated microRNAs in the INT-HA-induced M2-like macrophages and we explored the functions of these miRNAs in macrophage polarization. The enrichment results indicated that these miRNAs might participate in the process of macrophage polarization. Furthermore, the quantitative real-time polymerase chain reaction results showed that miR-935 may play an important role in M2 macrophage polarization which was activated by INT-HA.

microRNA expression in facet cartilage degeneration

(Submitter supplied) Osteoarthritis (OA) of spine (facet joints; FJ) is one of the major causes of severe low back pain and disability worldwide. The degeneration of facet cartilage is a hallmark of FJ OA. However, endogenous mechanisms that initiate degeneration of facet cartilage are unknown and there are no disease-modifying therapies to stop FJ OA. In this study we performed microRNA array analysis to identify differentially expressed microRNAs in facet cartilage from patients with FJ OA compared to facet cartilage from patients with lumbar disc herniation (LDH).

Rnase1 responsive miRNA

(Submitter supplied) To determine the importance of RNase1 on cellular miRNA function, we used a lentivirus system to stably express RNase1 in dicer wt and dicer ex5 RKO cells in serum free culture. RNase1 was found to govern miRNA that reside outside the pureview of Dicer. Assignment of RNase1 as miRNA-generating enzyme suggests that, like Dicer, RNaseA enzymes mediate essential physiological functions through regulatory RNA.
