The Hypoxia-regulated Ectonucleotidase CD73 is a Host Determinant of HIV Latency [RNA-seq]

(Submitter supplied) Deciphering the mechanisms underlying viral persistence is critical to achieving a cure for HIV infection. Here, we implement a systems approach to discover molecular signatures of HIV latently-infected CD4+ T cells, identifying the immunosuppressive, adenosine-producing ectonucleotidase CD73 as a key surface marker of latent cells. Hypoxic conditioning, reflecting the lymphoid tissue microenvironment, increases the frequency of CD73+ CD4+ T cells and promotes HIV latency. more...

Genomic annotation of a non-coding region deleted on 4q25 [CUT&RUN]

(Submitter supplied) we identified 7 families presenting a new cardiac entity associated with a heterozygous deletion of 2 CTCF binding sites on 4q25, inducing TAD fusion and chromatin conformation remodeling. Impact of ZAP-70 kinase inhibitors on gene expression in murine xenograft models of malignant T-cell lymphoma

(Submitter supplied) T-cell lymphoma (TCL) is a group of lymphomas derived from mature T and NK cells, often characterized by an aggressive clinical course with limited effective treatments available. Emerging evidence underscores the pivotal role of T cell receptor signaling (TCR) in the pathobiology of T cell lymphomas. ZAP-70, a protein tyrosine kinase belonging to the Syk / ZAP-70 family, is expressed in T lymphocytes and in NK cells and is an essential kinase in the proximal signaling of the TCR. more...

RGS1 Modulates Autophagic and Metabolic Programs and Is a Critical Mediator of Human Treg Function

(Submitter supplied) Regulatory T cells (Tregs) are critical mediators of immune tolerance and play a diametric role in cancer and autoimmunity. Tumor-infiltrating Tregs are often associated with poor prognosis in solid tumors, as their enrichment in the tumor microenvironment contributes to immunosuppression. Conversely, dysregulation in the Treg compartment can disrupt self-tolerance, leading to autoimmunity. Herein, we describe a novel regulator of Tregs, the GTPase activator, Regulator of G Protein 1 (RGS1), RGS1, demonstrating that RGS1-deficient human Tregs show downregulation of Treg-associated genes and are less immunosuppressive. more...

The methylome of buccal epithelial cells is influenced by age, sex, and physiological properties

(Submitter supplied) Epigenetic modifications, particularly DNA methylation, have emerged as regulators of gene expression and are implicated in various biological processes and disease states. Understanding the factors influencing the epigenome is essential for unravelling its complexity. In this study, we aimed to identify how the methylome of buccal epithelial cells, a non-invasive and easily accessible tissue, is associated with demographic and health-related variables commonly used in clinical settings, such as age, sex, blood immune composition, hemoglobin levels, and others. more...

An anciently diverged family of RNA binding proteins maintain correct splicing of ultra-long exons through cryptic splice site repression [iCLIP-seq]

(Submitter supplied) We previously showed that the germ cell specific nuclear protein RBMXL2 represses cryptic splicing patterns during meiosis and is required for male fertility. RBMXL2 evolved from the X-linked RBMX gene, which is silenced during meiosis due to sex chromosome inactivation. It has been unknown whether RBMXL2 provides a direct replacement for RBMX in meiosis, or whether RBMXL2 evolved to deal with the transcriptionally permissive environment of meiosis. more...

USF1 modulates transcription and cellular functions by regulating multiple transcription factors in Huh7 cells [ChIP-seq]

(Submitter supplied) Liver cancer, including hepatocellular carcinoma (HCC), is a malignant tumor type with high metastasis and mortality rate in worldwide. Upstream transcription factor 1 (USF1) is a canonical transcription factor (TF) and associated with the pathogenesis of several cancers, while its biological functions and molecular targets in HCC are not clear. In this study, we overexpressed USF1 in Huh7 cells and used whole transcriptome profile sequencing (RNA-seq) and chromatin immunoprecipitation sequencing (ChIP-seq) methods to systematically investigate the downstream targets of USF1. more...
