Quantitative Analysis of PPARD Transcriptomes in Colon Cancer Cells by Next Generation Sequencing (NGS)

(Submitter supplied) Purpose: NGS has revolutionized systems-based analysis of cell signaling pathways. The goal of this study is to determine the effects of PPARD in colon cancer cell transcriptomes in relation to the metastatic potential. Methods: NGS-derived colon cancer cell mRNA transcriptome profiles of HCT116 WT (HCT116) and HCT116 with genetic PPARD-knockout (KO1) cells were generated by deep sequencing, in quadruplicate, using Illumina HiSeq2000 .The transcriptomes of HCT116 and KO1 cells will be compared to determine the differentially expressed genes between HCT116 and KO1 cells. more...

PPARδ elicits ligand-independent repression of 2 Trefoil Factor Family to limit prostate cancer growth

(Submitter supplied) Cancer Res; 78(2); 399-409. ©2017 The nuclear receptor PPAR-β/δ (PPARD) has essential roles in fatty acid catabolism and energy homeostasis as well as cell differentiation, inflammation, and metabolism. However, its contributions to tumorigenesis are uncertain and have been disputed. Here, we provide evidence of tumor suppressive activity of PPARD in prostate cancer through a noncanonical and ligand-independent pathway. more...

Expression data from human lung cancer cell lines with NEDD9 overexpression or NEDD9 knockdown

(Submitter supplied) NEDD9 is important for lung cancer metastasis. However, the detailed mechanism remains elusive. Using the microarray data generated with human lung cancer cell lines with either NEDD9 overexpression or NEDD9 knockdown, we plan to idnetify important signal pathways regulated by NEDD9. This may explain how NEDD9 excutes its function in lung cancer. We used microarrays to detail the global programme of gene expression underlying cellularisation and identified distinct classes of up-regulated genes during this process.

Genome-wide analysis of mouse 4T1 tumor cells derived clone (4T1ch9) gene expression subjected to transduction with STC1-specific or non-targeting shRNA lentiviral particles

(Submitter supplied) Analysis of genes regulated by STC1 down-regulation in mouse 4T1 derived clone, 4T1ch9. STC1 expression is associated with tumor growth and metastasis. This study looks at genes affected when STC1 expression is down-regulated by STC1 shRNA.

Genome-wide analysis of colon cancer cell SW480_Control, _BOP1, _CKS2 and _NFIL3 treated with different inhibitors

(Submitter supplied) SW480 cells overexpressing BOP1, CKS2 or NFIL3 migrated more actively compared to Control cells. Migration induced by BOP1, CKS2 or NFIL3 was repressed by interfering with distinct signaling systems using small- molecular-weight inhibitors, i.e., interference with PI3K, JNK and Notch in the case of BOP1, with PI3K and p38 MAPK in the case of CKS2, as well as with PI3K, p38 and mTOR in the case of NFIL3. more...

A TGFbeta-PRMT5-MEP50 Axis Regulates Cancer Cell Invasion through Histone H3 and H4 Arginine Methylation Coupled Transcriptional Activation and Repression

(Submitter supplied) We sequenced mRNA from 3 biological replicates each of A549 lung adenocarcinoma cell lines expressing shRNA against GFP (control), PRMT5, or MEP50. We then determined differential gene expression.

The RhoA guanine nucleotide exchange factor Net1 is required for PyMT mediated mammary gland tumorigenesis and metastasis

(Submitter supplied) The Net1 RhoGEF has been implicated in invasive behavior in vitro, but its impact on tumorigenesis and metastasis in vivo has not yet been established. We developed Net1 knockout mice and profiled the transcriptomes of the resulting tumors to assess impact on pathway activation.

Gene expression profiling of FGFR1 KO mice

(Submitter supplied) A novel RCAS-Cre-IRES-PyMT (RCI-PyMT) virus was designed to specifically knockout genes of interest in tumors generated in appropriate mutant mouse hosts. We used this tumor knockout, or TuKO, strategy to concisely ablate fgfr1 in PyMT induced mammary tumors in K19-tva/fgfr1loxP/loxP mice. The similarly injected control K19-tva mice developed mammary tumors exhibiting high metastasis penetration to lung, making this an ideal model for breast cancer metastasis. more...

Claudin-1 is an Invasion and Metastasis Suppressor of Lung Adenocarcinoma

(Submitter supplied) Background: Claudin-1(CLDN1), one of the key components of tight junction proteins, was found to be down-regulated in human lung adenocarcinomas. This study we investigated the clinical significance of CLDN1 expression in lung adenocarcinoma patients and its role in cancer progression. Method: CLDN1 mRNA expression was measured in tumor specimens from 51 patients with lung adenocarcinoma. CLDN1 protein expression was also examined by immunohistochemistry on specimens from an independent cohort of 67 lung adenocarcinoma patients. more...

Claudin-1 overexpression effect on lung adenocarcinoma cell line

Analysis of lung adenocarcinoma CL1-5 cells overexpressing Claudin-1 (CLDN1), a component of tight junction complexes. Low CLDN1 expression in lung adenocarcinomas is associated with shorter overall survival. Results provide insight into the role of CLDN1 in the progression of lung adenocarcinoma.

Significant Downregulation of Platelet Gene Expression in Metastatic Lung Cancer

(Submitter supplied) Platelets have multiple roles in cancer cell metastasis. In this work we employed exon microarray technology to address platelet gene expression in metastatic non small cell lung cancer versus controls without cancer. We found that 197 of the 200 genes with the most significantly altered expression levels had their expression levels downregulated.
