SNP array analysis of neuroblastoma tumors

(Submitter supplied) Chromosome 1p LOH was seen in one-third of cases. LOH events on chromosomes 11q and 1p were generally accompanied by copy number loss. The one exception was on chromosome 11p, where LOH in all 4 cases was accompanied by normal copy number or diploidy, implying uniparental disomy. Amplification of MYCN was also noted, and also, amplification of a second gene, ALK, in a single case. Keywords: SNP array analysis

Lithium-responsive neurological pathway (kitam-affy-droso-427260)

(Submitter supplied) Lithium has been used for more than 50 years as the primary therapy for bipolar affective disorder (BPD) and is highly effective in its treatment. However, the molecular and cellular mechanisms underlying the lithium’s therapeutic action still remains elusive. The goal of the proposed study is to identify genes involved in the lithium-responsive neurological pathway using the fruit fly as a model organism. more...

Characterization of weight loss and weight regain mechanisms after Roux-en-Y gastric bypass in rats

(Submitter supplied) Roux-en-Y gastric bypass (RYGB) is the most effective therapy for morbid obesity, but it has a ~20% failure rate. We used our established RYGB model in diet-induced obese (DIO) Sprague-Dawley rats, which reproduces human bi-phasic body weight (BW) loss pattern, to determine mechanisms contributing to success (RGYB-S) or failed (RYGB-F) RYGB. DIO rats were randomized to RYGB, sham operated Obese, and sham operated obese pair fed-linked to RYGB (PF) groups. more...

Genome-wide gene expression profiling of rice stigma

(Submitter supplied) In angiosperms, stigma provides initial nutrients and guidance cues for pollen grain germination and tube growth. However, little is known about genes that regulate these processes in rice. Here we generate rice stigma-specific gene expression profiles through comparing genome-wide expression patterns of hand dissected unpollinated stigma at anthesis with seven tissues including seedling shoot, seedling root, mature anther, ovary at anthesis, seeds of five days after pollination, 10-day-old embryo, 10-day-old endosperm as well as suspension cultured cells by using 57K Affymetrix rice whole genome array. more...

Inflammatory response to titanium dioxide particles exposure is enhanced during pregnancy

(Submitter supplied) Rationale: Maternal immune responses can promote allergy development in offspring. Pilot data show that neonates of mother mice exposed during pregnancy to air pollution particles have increased allergic susceptibility. Objective: We investigated whether inflammatory response to titanium dioxide (TiO2) particles earlier considered immunologically ‘inert’ is enhanced during pregnancy. Methods: Pregnant BALB/c mice (or non-pregnant controls) received particle suspensions intranasally at day 14 of pregnancy. more...

Expression data from Mus musculus Myc-null heart at E13.5

(Submitter supplied) We created mice, which are deficient for Myc specifically in cardiac myocytes by crossing crossed Myc-floxed mice (Mycfl/fl) and MLC-2VCre/+ mice. Serial analysis of earlier stages of gestation revealed that Myc-deficient mice died prematurely at E13.5-14.5. Morphological analyses of E13.5 Myc-null embryos showed normal ventricular size and structure; however, decreased cardiac myocyte proliferation and increased apoptosis was observed. more...

Sensitive expression profiling of T-cell subsets reveals parallel post-thymic differentiation for CD4+ and CD8+ lineages

(Submitter supplied) Gene expression profiling has opened new directions of investigation in scientific research, however, the need for substantial amount of biological material to perform these experiments limits significantly its application range. Here, we have developed procedures to perform microarray analysis on amplified cDNA from single cells, including primary T lymphocytes, and applied this technology to the study of CD4 and CD8 lineage differentiation. more...

Bovine ovarian follilce selection

(Submitter supplied) Lack of, or abnormal, ovarian follicular development prevents establishment of pregnancy in human and is associated with reduced reproductive efficiency in farm animals. Development of new treatments is limited by our poor understanding of ovarian follicular development. The objective of this study is to identify changes in gene expression during the selection stage of bovine follicular growth. Follicles of different sizes were collected and intrafollicular concentrations of steroids were measured to identify healthy follicles. more...

Profiling of mucosal derived bovine ab and gd T cells during Salmonella enterica serovar typhimurium enterocolitis

To better understand the precise roles of gd T cells relative to ab T cells in a specific infection, we utilized Salmonella Enterica Serovar Typhimurium (S. typhimurium) infection in cattle as it is a leading cause of disease in cattle and closely approximates S. more...

Repression of pleiotrophic drug resistance genes in Saccharomyces cerevisiae using chimeric transcriptional repressors

(Submitter supplied) Yeast is a powerful model system for studying the action of small molecule therapeutics. An important limitation has been low efficacy of many small molecules in yeast due to limited intracellular drug accumulation. We used the DNA binding domain of the pleiotropic drug resistance regulator Pdr1 fused in-frame to transcription repressors to repress Pdr1 regulated genes. Expression of these regulators conferred dominant enhancement of drug sensitivity and led to greatly diminished levels of Pdr1p regulated transcripts, including the yeast p-glycoprotein homologue Pdr5. more...

Cytotoxic activity of HTI-286 in prostate cancer

(Submitter supplied) HTI-286, an analogue of hemiasterlin, interferes with microtubule dynamics and circumvents transport-based resistance to taxanes. In this study we evaluate the inhibitory effects of HTI-286 on human prostate cancer growth in vitro and in different in vivo models. The results show that HTI-286 is a potent inhibitor of proliferation and induced marked increases in apoptotic rates in all cell lines tested. more...

Differences found in expression of cell surface & ECM components during development of the rainbow trout ovary & testis.

(Submitter supplied) A comparison of "maturing" and "prespawn" ovarian and testicular transciptomes was performed to determine the genes that are involved in regulating gametic and accessory cell function during maturation and development of the rainbow trout gonad. To identify some fo the genes involved in these processee, total RNA was compared between three-year-old normal vs two-year-old normal (maturing) and three-year-old normal vs two-year-old precocious (prespawn) gonadal tissue. more...

Correlation analysis of genome-wide gene expression profiles by DNA microarray

(Submitter supplied) We previously isolated a new angiogenesis inhibitor, terpestacin, from natural products. To investigate molecular mechanism of terpestacin for its anti-angiogenic activity, we identified a cellular binding protein of terpestacin using phage display biopanning analysis. Terpestacin specifically bound to the small molecular ubiquinone binding protein (QP-C), a 13.4 kD subunit of the mitochondrial Complex III. more...

Redirection of metabolism for biological hydrogen production

(Submitter supplied) Photosynthetic microbes can produce the clean-burning fuel hydrogen using one of nature’s most plentiful resources, sunlight 1,2. Anoxygenic photosynthetic bacteria generate hydrogen and ammonia during a process known as biological nitrogen fixation. This reaction is catalyzed by the enzyme nitrogenase and consumes nitrogen gas, ATP and electrons 3. One bacterium, Rhodopseudomonas palustris, has a remarkable ability to obtain electrons from green plant-derived material 4,5 and to efficiently absorb both high and low intensity light energy to form ATP 6. more...
