Summary
This genomic region represents a DNase I hypersensitive site (DHS) that was predicted to be an enhancer by the ENCODE (ENCyclopedia Of DNA Elements) project based on various combinations of H3K27 acetylation and binding of p300, GATA1 and RNA polymerase II in K562 erythroleukemia cells. It was validated as an active enhancer by reporter assays in K562 erythroleukemia cells. Multiple overlapping subregions were validated as cis-regulatory elements by CRISPR/Cas9-mediated perturbation in K562 cells, where one subregion was shown to regulate the GATA1 (GATA binding protein 1) gene with high confidence and the CCDC22 (coiled-coil domain containing 22) gene with lower confidence, another subregion was validated as a positively-acting regulatory element for GATA1 and a negatively-acting regulatory element for PLP2 (proteolipid protein 2), while another subregion was shown to be a regulatory element for both GATA1 and HDAC6 (histone deacetylase 6).
