Study has demonstrated that ARX is a novel substrate of PRKCA, with phosphorylation at serine 174, and that the c-terminal region of ARX required to bind PICK1. Binding causes a shift in PICK1 subcellular localization to the nucleus to co-locate with the ARX protein, and truncation of this C-terminal region leads to the same loss of transcriptional activation as S174A mutant.

This study defines a novel mechanism whereby elevated [Ca(2+)] induced by NMDA receptor activation modulates Ago2 and miRNA activity via PICK1. Our work suggests a Ca(2+)-dependent process to regulate miRNA activity in neurons in response to the induction of long-term depression.

The PICK1 Ca(2+) sensor modulates N-methyl-d-aspartate (NMDA) receptor-dependent microRNA-mediated translational repression in neurons.
