Manufacture protocol The 6K Yeast Genome Array contains 6389 70mer probes representing 6389 Saccharomyces cerevisiae open reading frames (ORFs). All 6389 probes are designed from the set of open reading frames (ORFs) obtained from the Saccharomyces Genome Database (SGD) at Stanford University (http://genome-www.stanford.edu/Saccharomyces/). The cDNA library is arrayed in 75×25 mm、chemically modified glass slide using SmartArrayTM (CapitalBio Corp., Beijing, China ). The yeast housekeeping gne Actin、RPS4B and yeast genomic DNA (used as positive control) were also arrayed in the same glass slide. Hex was used as positive control in the pointting step, while salmon sperm DNA and 50% DMSO buffer was used as negative control in the pointting step. All the dot matrix divided into 48 sub-matrixes. The dot spacing is 200 um and the dot diameter is 160 um. Each gene repeat three times.
