Description A collection of 2034 human genes corresponding to specific key-words was selected by direct interrogation of UniGene database. Primer pairs for the amplification of these 2304 human ORFs were obtained using the Primer 0.5 software. Specificity of each primer sequence was verified by blast analysis. The primers were designed to amplify a 400-600 bp product for each ORF. PCR amplification of each gene was performed using a mix of cDNAs obtained from various cell lines as template.
The PCR products were prepared in 96-well plates, purified by ethanol precipitation, washed in 70% ethanol, dried, dissolved in TE/DMSO (50/50) and stored at -80°C. PCR products were arrayed on amino-silane coated glass slide (Corning CMT-GAPS II) using a MicroGrid-II Micro-arrayer (BioRobotics, Cambridge, UK).
