Manufacture protocol The 32,450 BAC clones were amplified using three independent DOP PCR amplifications, followed by pooling of the DOP PCR products and re-amplification using a universal amino-modified primer. The amino-PCR-products were then purified using Millipore filter plates, eluted with printing buffer. Quality control agarose gel electrophoresis was performed for all clones after the universal amino PCR, and the gel images were stored in an image database. All DNA transfers were performed using a Hydra 96 channel micro-dispenser (Matrix Technologies, Hudson, NH) and a Biomek FX robot (Beckman-Coulter, Fullerton, CA). The aminogroup attached at the 5’ end of the universal primer, allows for covalent attachment of DNA fragments to the CodeLink HD slide surface (GE Healthcare, Piscataway, NJ). We used the spotting solution recommended for these slides (GE Healthcare). Printing of the clones was carried out using a high throughput printing robot (http://www.heflingenetics.uab.edu/CORE/array_printing.shtml). The center-to-center distance between the spots was 160 micrometers using a hexagonal printing pattern.
