Title A pooled single-cell genetic screen identifies regulatory barriers in the continuum of the epithelial-to-mesenchymal transition
Organism Homo sapiens
Experiment type Expression profiling by high throughput sequencing
Summary We demonstrate that progression along EMT occurs along a continuum and that interruption of key signaling events reveals regulatory “barriers” that mimic discrete stages.

Overall design Single-cell RNA-seq experiments were performed on the 10X Genomics platform for MCF10A and HuMEC cells plated and harvested as a function of space. These experiments were repeated in MCF10A cells expressing Cas9 and transduced with a CROP-seq library targeting EMT-associated cell surface receptors and transcription factors.
