Overall design |
C57BL/6J mice were fed High Fat Diet (Research Diet, D12331) and control diet (Research Diet, D12329) for 12 weeks. Mice were sacrificed by cervical dislocation at ZT0 (6 am) and at ZT12 (6 pm), liver collected and snap frozen. ChIP was performed as previously described (Uhlenhaut et al. 2012). Sheared chromatin was immunoprecipitated with anti-H3K27ac antibody (Abcam, ab4729). Two replicates per condition and time point were used, for a total of 8 ChIP-seq samples.
