Title Comparison of tumor-associated YAP1 fusions identifies a recurrent set of functions critical for oncogenesis[RNASeq]
Organism Homo sapiens
Experiment type Expression profiling by high throughput sequencing
Summary Several different YAP1 gene fusions have been identified in various human cancers. We performed RNA sequencing on human neural stem cells overexpressing either YAP1-MAMLD1, YAP1-FAM118B, or YAP1-TFE3, as well as on whole tumor lysates from mouse brain tumors generated by intra-cranial expression of either YAP1-MAMLD1, YAP1-FAM118B, or YAP1-TFE3.

Overall design We performed RNA sequencing on human neural stem cells overexpressing either YAP1-MAMLD1, YAP1-FAM118B, or YAP1-TFE3. U5 huamn neural stem cells were transduced with pLJM1-"transgene"-PGK-Puro. 48 hrs post transduction, positive clones were selected by 72 hrs of Puromycin selection. Total RNA was extracted 48 hrs after Puromycin selection.
