Summary ABSCISIC ACID INSENSITIVE3 (ABI3) is a transcription factor that is a member of the B3 domain family and is involved in seed development. To understand how it controls development, we mapped genome-wide binding sites by ChIP-chip and combined this data with transcriptomic data obtained by microarrays.

Overall design ABI3 was expressed by its native promoter and included a C-terminal 10x c-myc tag. This transgene was able to rescue the abi3-5 mutant. A second transgene (35S:AGL15) allowed establishment of stable somatic embryo cultures. ChIP was performed using anti-c-myc antibody (Myc-Tag (9B11) Mouse mAb; Cell Signaling 2276S) on tissue expressing ABI3-myc (three biological replicates) and on three controls of tissue without c-myc antibody.
