Summary |
During repair phase after naphathalene injury 3 subsets of myeloid cells accumulate at the site of injury including P1 (monoyte), P2 (monocyte-derived macrophages and P3 (resident alternatively activated airway macrophages). ST2 receptor was found to be expressed by P2 and P3 subsets. To understand which biological processes in lung macrophages were regulated by ST2, myeloid cell subsets were flow-sorted from Naphthalene-treated WT and ST2-/- mice on d6 and their transcriptome analyzed using RNA-seq

Overall design |
ST2Ko and C57Bl/6J female mice (9-10 weeks) were treatment with naphthalene at a dose of 200-250 mg/kg. On day 6, mice were euthanized and their lungs were collected and digested for myeloid cell sorting of P1 (CD11b+ ly6c+ cells), P2 (CD11b+ CD11c+ F4/80+ cells) and P3 (Siglrc-F+ CD206+ CD11c+ F4/80+ cells). then P1-P3 were RNAseq analysis.
