Summary |
We analyzed the RNA expression pattern in B3GALT5-Knockout H9 cells (B3GALT5-KO) relative to wild type H9 cells (WT) by RNA-seq. Collectively, our results reveal that B3GALT5-Knockout cells cultured in 2iLAF condition represent naïve-specific characteristics as well as WT cells convert to naïve state in 5iLAF condition. These findings support that loss of B3GALT5 which result in depletion of primed-specific GSLs not only do not have influence on maintain pluripotency and ability to specific lineage differentiation but also could promotes naive pluripotency generation in hESCs.

Overall design |
This data includes five RNA-seq datasets. Two primed, two naïve hESCs and 1 control hESC samples were profiled in experiments. Primed samples consist of H9 hESCs with or without knockout of B3GALT5 where indicated, in standard primed hESC conditions. Naïve samples consist of H9 hESCs with or without knockout of B3GALT5 that were profiled after conversion to the naïve state (5iLAF or 2iLAF medium where indicated). A control sample is WT of H9 cultured in 2iLAF medium.
