Summary |
To get a genome wide picture of the effects of TOR knockdown during the early phase of lateral root formation, we compared by RNA-seq the transcriptomes of roots 6h after the synchronous induction of lateral formation by auxin treatment (Himanen et al, 2002 - http://dx.doi.org/10.1105/tpc.004960) in the inducible UB10pro>>amiR-TOR background.

Overall design |
Samples (Col-0 or inducible UB10pro>>amiR-TOR line) were prepared for harvesting using the synchronous induction of lateral root procedure. All samples were pre-treated with 10 µM NPA for 24 h then shifted to plates containing 10 µM NPA and 10 µM Estradiol or DMSO control for additional 24 h before being shifted to either 10 µM IAA + 10 µM Estradiol or DMSO for LR induction or maintained on the same plates. Root tissue was harvested after 6 h. All sampling points were performed in triplicate. For each sample, about 200 segments of the lower two-thirds of the seedling roots were pooled.
