Summary |
Purpose: mRNAs sequencing of the gene expression differences in diffDC transfected with si-nc or si-Suclg2, the goals of this study are to investigate the biological effect of the metabolic enzyme succinate-CoA ligase subunit beta Suclg2 in regulating regulatory DC differentiation and function.

Overall design |
Bone marrow cells from C57/BL6 mice were cultured with GM-CSF and IL-4 for 6 days and stimulated with LPS for 24 hours to generate bone-marrow derived DC (BMDC). CD11c+ BMDC were isolated by anti-mouse CD11c magbeads to generate maDC. maDC were then cocultured with spleen stroma cells for 9 days to generate diffDC. And then diffDCs were transfected with si-Suclg2 or si-nc. diffDCs transfected with si-Suclg2 or si-nc were subjected to RNA seq.
