Title Cytokines in the ApoE-/- mice atherosclerotic plaque

Summary To identify the dynamic changes of cytokines in the ApoE-/- mice atherosclerotic plaque, heart, and serum upon β-adrenergic stimulation (isoproterenol, 5 mg/kg body weight) at 0.5hrs, 1hrs, 3hrs and 24hrs using mouse cytokine antibody arrays.
 
Overall design 45 samples. Male ApoE knockout mice (8 weeks of age, C57BL/6J background) fed a high fat diet containing 40 kcal% fat and 1.25% cholesterol (D12108C, Reaserch Diets, USA) for 2 weeks. Carotid collar placement was performed 2 weeks later to induce carotid artery plaque. After carotid collar placement and high-fat feeding for 3 months, mice were subcutaneously injected with a single dose of ISO (Sigma-Aldrich, St. Louis, MO, USA; 5 mg/kg body weight, as determined effective by dose-response preliminary experiments) or saline (10 mL/kg body weight).There are 9 groups: vehicle group, mice were subcutaneous injected with a single dose of saline (10 ml/kg body weight), ISO groups, mice were subcutaneous injected with a single dose of isoproterenol (5 mg/kg body weight) after 0.5hrs, 1hrs, 3hrs and 24hrs (vessel, heart and plasma groups by different pionts)
