Title A2AR-eGFP reporter mouse enables elucidation of A2AR expression dynamics during anti-tumor immune responses

Summary Understanding of the mechanism by which A2AR is regulated has been limited by difficulty in identifying the cell types that express A2AR due to a lack of robust antibodies for these receptors. To overcome this we developed an A2AR-eGFP reporter mouse and assessed the expression of A2AR during ongoing anti-tumor immune responses.
Overall design A2AR eGFP reporter mice were injected with 5 x 10^5 AT-3 ova tumors subcutaneously and where indicated treated at days 14 and 18 post tumor inoculation with anti-PD-L1. At day 21 post tumor inoculation CD8+ GFP+ or GFP- tumor-infiltrating lymphocytes were flow sorted and analyzed by 3’RNA-Sequencing.
