Summary |
In situ transposition followed by laser capture microdissection was applied to spleen mouse tissue sections to demonstrate feasibility of LCM-based spatial chromatin accessibility analysis of regions of interest. B220 and CD3e immunostained B and T cell zones were analyzed.

Overall design |
Fresh frozen spleen tissue sections from C57BL/6J WT mice were tagmented in situ to assess chromatin accessibility. LCM was employed to collect a mini bulk of immunostained B and T cells. Two mini-bulk sizes were investigated: 100 single-captured nuclei or 10 larger captured-areas (patches) composed each of roughly 100 nuclei, for a total of ~1000 nuclei. Four technical replicates were collected at least per condition.
