Overall design |
Mobilized apheresis products were collected from de-identified, healthy donors. Isolated CD34+ HSPCs were cultured in myeloid expansion medium for 10 days with or without TNFα supplement. For bulk ATAC sequencing, CD14+ monocytic cells were further isolated at day 10. The cells were then thawed in a 37oC water bath, pelleted, washed with cold PBS, and tagmented as previously described (Buenrostro et al. 2013), with some modifications based on (Corces et al. 2017).
