Sample GSM1619694 Query DataSets for GSM1619694
Status Public on Feb 24, 2016
Title KBC11_miRNA-4_0
Sample type RNA

Source name superficial temporal artery tissue
Organism Homo sapiens
Characteristics subject status: patients undergoing craniotomy for cerebral aneurysms
subject/sample id: KBC11
tissue: superficial temporal artery (STA) tissue
Growth protocol Human samples of cerebral aneurysm and superficial temporal arteries were extracted surgically from patients undergoing craniotomy for cerebral aneurysms.
Extracted molecule total RNA
Extraction protocol After successful clipping of the aneurysm, the dome was resected with the use of micro-scissors and bayonetted forceps (not used before during the case) and was placed immediately into a vial containing RNAlater solution (Ambion, Austin, TX) to protect RNA from degradation. The vials were immediately frozen to -80 °C. Small RNA was then size selected with mirVana miRNA isolation kit.
Label biotin
Label protocol Following the manufacturer’s protocol, 130ng of total RNA was labeled with the FlashTag Biotin HSR RNA labeling kit (Affymetrix, Santa Clara, CA). To confirm efficient labeling the ELOSA QC assay (Affymetrix, Santa Clara, CA) was performed.

Hybridization protocol Samples were hybridized on the Affymetrix 4.0 microarrays overnight.
Scan protocol Washing, staining and scanning of the arrays was performed using the Command Console, and the spike-in analysis of control oligonucleotides was evaluated using the Expression Console.
Description Affymetrix GeneChip miRNA array 4.0 for Superficial temporal artery sample KBC11
Data processing Data were analyzed using the recommended Transcriptome Analysis Console (TAC) software from Affymetrix. Only human microRNAs were included in the analysis, resulting in reduced data set of 6,631 rows.
