Hybridization protocol |
Plasma hybridization was performed on TECAN HS 4800 Pro. 1. Protein arrays were blocked with 5% milk PBST (0.2% Tween) for 1 hour at RT; 2. 150ul 1:50 diluted serum was applied to each slide with medium agitation 16 hours at 4oC; 3. Slides were incubated with 150ul 1:500 dilution of Dylight 649-linked anti-human secondary antibodies (Jackson Immunoresearch) in milk PBST.

Scan protocol |
Slides were scanned by TECAN PowerScanner at 50% gain, 50% laser and 10µm resolution.

The serum was applied on the protein array, secondary anti-human IgG antibody used for detection.

Data processing |
1. The background signal estimated by the 25th percentile of the negative control; 2. Normalize the array by the median.
