Sample GSM2198067 Query DataSets for GSM2198067
Status Public on Apr 17, 2017
Title TL28
Sample type SRA
Source name H1 POU5F1-IRES-eGFP
Organism Homo sapiens
Characteristics cell type: H1 POU5F2-eGFP reporter cells
Treatment protocol Regular hESC culture without any special treatment.
Growth protocol Cells were cultured at 37 celsius degree in 5% CO2, in Essential 8 media (Stemcell Technologies).
Extracted molecule genomic DNA
Extraction protocol 4C-Seq library was prepared following the protocol described in van de Werken et al. 2012 (PMID: 22961246). DpnII and NlaIII were used as the 4 base cutter in 4C-Seq.
Libraries were prepared according to Illumina's instructions, using TruSeq DNA Sample Preparation Kits v2, Set A (Cat# FC-121-2001). 4-12 library preps were mixed for multiplexed single-read sequencing using Illumina Hi-seq 2000 (Illumina).

Instrument model Illumina HiSeq 4000
Description eGFP-/POU5F+ cells, sorted from H1 POU5F1-eGFP reporter cells after CREST-seq screen, replicate 2
Data processing Library strategy: 4C-Seq
4C-Seq data were mapped using 4Cseqpipe (van de Werken et al., 2012) with the first cutter set as DpnII and the second cutter set as NlaIII.
