Treatment protocol |
Using RNAiMAX (Invitrogen) reagents, Huh-1 cells were reverse-transfected with control siRNA or siLSD1. After 48hrs, cells were incubated under 1% O2 for 24hrs.

Growth protocol |
Huh-1 cells were maintained in Dulbecco’s modified Eagle medium (DMEM; Gibco BRL, Grand Island, NY). Media was supplemented with heat-inactivated 10% fetal bovine serum (GIBCO BRL) and 1× Antibiotic-Antimycotic (GIBCO BRL). Cells were cultured at 37℃ in a humidified incubator under 5% CO2. Hypoxic culture conditions were constructed with O2/CO2 incubator (1%O2, 5% CO2 and 92%-94% N2, MCO-5M, Sanyo, Japan)

Extraction protocol |
Total RNA extraction was perforemd using QIAGEN RNeasy mini kit according to manufacturer's instruction. Total RNA integrity was determined with ExperionTM RNA StdSens (BIO-RAD).

Illumina TotalPrep RNA Amplification kit (Applied Biosystem/Ambion, Austin, TX, USA) was used to generate biotin-UTP labeled cRNA according to manufacturer’s instructions

Labeled cRNA (750ng) was hybridized to HumanHT-12 v4 chips at 58℃ for 18 hours.

Scan protocol |
After hybridizing procedures, chips were scanned using a BeadArray Reader and BeadScan software (Illumina).
