Homo sapiens |
Characteristics |
cell type: peripheral blood mononuclear cell treatment response: responder timepoint: -3 days treatment: None
Treatment protocol |
RNA from peripheral blood mononuclear cell of 8 patients at 2 different time points (3 days before and 6 days after Sorafenib treatment) were extractied from 8 ml of blood.
Extracted molecule |
total RNA |
Extraction protocol |
2ug of total RNA from about 10^7 cells per sample were incubated with DNaseI according the Qiagen manufacture and cleaned up with RNeasy Micro Kit (Qiagen, Hilden, Germany).
Label |
Biotin
Label protocol |
RNA integrity was analyzed by capillary electrophoresis using a Fragment Analyser (Advanced Analytical Technologies, Inc. Ames, IA) and were further processed with the Affymetrix GeneChip Pico kit
Hybridization protocol |
Total RNA was hybridized to Affymetrix Clariom S arrays as described by the manufacturers (Affymetrix/Thermo Fisher Scientific, 168 Third Avenue, Waltham, MA USA 02451.)
Scan protocol |
Arrays were washed and stained with GeneChip Fluidics Station 450 and scanned with GeneChip Scanner 3000 using the GeneChip Operating Software (Affymetrix).
