Treatment protocol Plant was grown in iron sufficient (100 uM Fe(NO3)3) hydroponics for 14 days prior to harvest of 2nd trifoliate.
Growth protocol Seedlings were germinated in sterile vermiculite. Seven days after planting, seedlings were transplanted to iron limited hydroponics system. A pH of 7.8 was maintained by an aeration of 3% CO2:Air mixture throughout the hydroponics system. Plants were grown in hydroponics until 2nd trifoliate stage, approximately 14 days.
Extracted molecule total RNA
Extraction protocol RNA was extracted from leaf tissue using Qiagen's RNeasy RNA extraction kit following the protocol provided by the manufacturer. RNA integrity was evaluated on an Agilent Bioanalyzer 2100 using an Agilent RNA Nano LabChip (Agilent technologies, Palo Alto, CA)
Label Biotin
Label protocol Samples were labeled with the Affymetrix One Cycle labeling kit, following manufacturer's directions
 
Hybridization protocol Biotin labeled cRNA was hybridized to the Affymetrix soybean genome array as reccomended by Affymetrix (Affymetrix GeneChip Analysis Technical Manual; Affymetrix Inc, Santa Clara, CA) at the iowa State University Gene Chip Facility (http://www.biotech.iastate.edu/facilities/Genechip.htm)
Scan protocol GeneChips were scanned using the Affymetrix GeneChip Scanner 3000 at the Iowa State University Gene Chip Facility (http://www.biotech.iastate.edu/facilities/Genechip.htm).
Description Data was collected from the 2nd trifoliate of Soybean genotypes which differ in their iron stress response grown under either iron sufficient or deficient conditions.
Data processing Results were analyzed usinig the GeneTraffic Microarray Analysis System. An MBEI transformation of the data was performed according to internal GeneTraffic algorithms.
