Extraction protocol Single cell was randomly picked with mouth pipette, then washed by 1mg/ml HSA() and transferred into prepared lysis buffer. The cells can store at -80℃ or do it immediately. We used a modified SMART-seq2 protocol to lysis for release all RNA, reverse transcription for capture the cDNA of mRNA and pre-amplification for increase yields of cDNAs. The resulting cDNAs from single cells were barcoded, and then pooled together with different barcode. Followed by 4 cycles, the index of sequence with biotin modified was added at 3’ end.

Description single cell RNA seq of 48 heart tissue cells
Data processing Illumina CASAVA version 1.8 were used to the basecalling.
Sequencing adapters, low quality sequences and amplification primers were trimmed from the raw sequencing reads.
For RNA-seq data, trimmed clean reads were aligned to human reference genome (hg19) using TopHat (v 2.0.12) -pass mapping steps.
