Single cell was randomly picked with mouth pipette, then washed by 1mg/ml HSA() and transferred into prepared lysis buffer. The cells can store at -80℃ or do it immediately. We used a modified SMART-seq2 protocol to lysis for release all RNA, reverse transcription for capture the cDNA of mRNA and pre-amplification for increase yields of cDNAs. The resulting cDNAs from single cells were barcoded, and then pooled together with different barcode. Followed by 4 cycles, the index of sequence with biotin modified was added at 3’ end.

Data processing |
Illumina CASAVA version 1.8 were used to the basecalling. Sequencing adapters, low quality sequences and amplification primers were trimmed from the raw sequencing reads. For RNA-seq data, trimmed clean reads were aligned to human reference genome (hg19) using TopHat (v 2.0.12) -pass mapping steps. Genome_build: hg19 Supplementary_files_format_and_content: TPM.txt files include the TPM values of RefSeq genes
