Total RNA was isolated from early passages of tranfected cells cell lines (referred to as “Huh7-S33Y” and “Huh7-Vec” based on construct integrated into genome of Huh7) using the guanidine isothiocyanate method.

Description |
We performed SAGE experiments with Invitrogen's I-SAGE kit (Invitrogen, San Diego, CA USA) according to the manufacturer’s instructions and EMBO’s SAGE manual, by using 25 ug of total RNA from both samples. The kit manual was followed until preparation of 100 bp ditags. After this step, some minor modifications were made as described in the EMBO’s SAGE manual. Sequencing was performed using an Amersham 96 capillary automatic DNA sequencer.
