Hybridization protocol |
Genomic DNA from cowpea and adzuki bean were mixed with SeqCap EZ Developer Reagent. The SeqCap EZ Developer Reagent is an alternative of human Cot-1 DNA specifically designed for plants to minimize non-specific binding of probes and DNA. After being denatured at 95°C, the mixture was applied to the microarray, and the hybridization was carried out at 57°C for 40 hours (Oligo aCGH hybridization kit, Agilent Technologies).

Scan protocol |
After hybridization, the microarray was washed with OligoaCGH wash buffer at 37°C, and slides were then dried. The arrays were then scanned using an Agilent Scanner (Agilent Technologies)
