Treatment protocol |
Embryos were dechorionated with 50% bleach, put on a cover slip and covered with Halocarbon oil 27 (Sigma). Embryos of the appropriate stage were manually selected under the dissecting scope. Selected embryos were transferred to a basket, rinsed with PBS with 0,7% NaCl, 0,04% triton-X100 and placed on ice in the Trizol solution (GibcoBRL).

Growth protocol |
db/db mice received a single 200 microliter intravenous injection of 1E11 vector genome adeno-associated virus (AAV) encoding FGF19 or green fluorescent protein via tail vein. Animals were euthanized and livers were collected 2 weeks after injection of the AAV vectors for gene expression analysis.

Extraction protocol |
Mouse livers were snap-frozen in liquid nitrogen upon euthanization of animals. Total RNA was extracted using RNeasy Mini kit (Qiagen) and treated with DNase I according to the manufacturer's instructions.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 1 ug total RNA (GeneChip WT cDNA synthesis and amplification kit and GeneChip WT terminal labeling kit from Affymetrix).

Hybridization protocol |
Following fragmentation, cRNA were hybridized for 16 hr at 45C on Affymetrix mouse gene 1.0 ST whole-transcript Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.

Scan protocol |
GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G2500A.
