RNA |
Source name |
Control RNAi in whole body
Organism |
Caenorhabditis elegans |
Characteristics |
strain/background: N2 tissue: whole body RNAi: Control
Treatment protocol |
N2, VP303, NR350 or TU3401 worms were subjected to control or ash-2 RNAi for 3 days from egg stage to young adult stage.
Growth protocol |
All strains were maintained at 20 °C on nematode growth medium (NGM) plates seeded with Escherichia coli OP50 as previously described.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was isolated using TRIzol reagent (Invitrogen) in N2, VP303, NR350, TU3401 worms under Control RNAi and ash-2 RNAi conditions.
Label |
biotin
Label protocol |
Ambion WT cDNA Expression Kit was used according to the protocol.
Hybridization protocol |
Fragmented and labelled cDNA was hybridised to GeneChip C. elegans Gene 1.0 ST Array (Affymetrix).
Scan protocol |
Arrays were scanned using the Affymetrix GeneChip Scanner 3000.
Data processing |
The probe set signals were calculated with the robust multi-array average (RMA) algorithm implemented in GeneSpring GX 11.0.2 (Agilent Technologies).
