Treatment protocol |
An exercise session (60 min, one-legged knee extension, ~65% of maximal power in the one-legged ramp test) started 2 h 35 min after a standardized breakfast. The participants ate a standardized lunch 1 h 20 min after the exercise session. This test session was repeated after an aerobic training programme (2 month, 1 h/day, 5 sessions/week) comprised training sessions on cycle ergometer (60 min) that alternated between continuous and intermittent exercise.

Samples from the vastus lateralis muscle were taken using a microbiopsy technique and were frozen in liquid nitrogen; RNA was extracted from the samples using an RNeasy mini kit (Qiagen) with DNase treatment (Fermentas, Lithuania) Libraries were constructed from 300 ng of RNA using a NEB Next Ultra II RNA kit. Poly-A mRNA was isolated using magnetic particles associated with 24-dT oligonucleotides by means of NEB Next Poly(A) mRNA Magnetic Isolation Module.
