Total RNA was isolated using Trizol reagent (Molecular lab), followed by clean up with RNeasy® Mini kit (Qiagen, Valencia, CA). RNA quality and integrity were confirmed by Bioanalyzer (Agilent technology)

cRNA samples were prepared for hybridization according to Illumina's instructions: 500 ng of total RNA from each sample was converted to cDNA synthesis then to biotin labeled cRNA by in vitro transcription using the Illumina TotalPrep RNA Amplification kit (Ambion Inc., Austin, TX). cRNA concentrations were measured using spectrophotometer (Nanodrop, ND-1000) and cRNA size was measured using Agilent bioanalyzer.

Hybridization protocol |
1.5 ug/array of biotin-labled cRNA was hybridized to Illumina Human-6 v 2 (48K) according to the Manufacturer's instructions (Illumina, Inc., San Diego, CA). Amersham fluorolink streptavidin-Cy3 (GE Healthcare Bio-Sciences, Little Chalfont, UK) was used to develop the array signal.
