Status Public on Sep 30, 2020
Title B_d2_1
Sample type SRA

Source name MEF
Organism Mus musculus
Characteristics treatment: infected with Brn3b-lentiviruses
collecting day: day 2
Treatment protocol MEFs were infected with Ascl1/Brn3b/Isl1 overexpressing lentiviruses, doxycycline was added on day 0 to induce Ascl1/Brn3b/Isl1 expression, cells were switched to neuronal medium (DMEM/F12+GlutaMAX+N2+B27+Forskolin+FGF2) on day 2 of reprogramming; medium was half-changed every other day.
Growth protocol MEFs were cultured in DMEM+10% FBS, at 37℃ in incubator.
Extracted molecule total RNA
Extraction protocol TRIzol following manufacturer's protocol
NEBNext UltraTM RNA library Prep Kit for Illumina following manufacturer's protocol

Library strategy RNA-Seq
Library source transcriptomic
Library selection cDNA
Instrument model Illumina HiSeq 2000

Description d2 and d7-gene counts
Data processing Basecalls performed using CASAVA
clean reads were aligned to the mouse reference genome (mm10) using Hisat2
featureCounts was used to count the reads numbers mapped to each genes
