Treatment protocol After maturation of macrophage precursors, the cells were washed and incubated for 4 hours with or without L. major promoastigotes. The cells were then washed free of unbound parasites and incubated for 24hours before harvesting for RNA preparation.
Growth protocol Bone marrow-derived macrophages from 6 mice each of C57BL/6, BALB/c, C.B6-(lmr1, lmr2) and B6.c-(lmr1, lmr2) were cultured for 3 days in the presence of FCS and L-cell conditioned media. After 2 days, adherent cells were discarded. This served to removed any fibroblasts present in the culture. The remaining cells were cultured for a further 4 days allowing non-adherent macrophage precursors to mature and adhere to the culture dish.
Extracted molecule total RNA
Extraction protocol RNA was prepared by using RNeasy (Qiagen, Valencia, CA).
Label biotin
Label protocol Samples were labelled using the Enzo BioArray HighYield RNA Transcript Labeling kit – Affymetrix, following the manufacturer's protocol.
 
Hybridization protocol Samples were hybridised using Affymetrix protocols.
Scan protocol GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G2500A.

Data processing The data were background corrected, normalized and summarized with the Bioconductor package gcrma. Full (fast=FALSE) parameter estimation was used.
