Title |
tumor 2 from male M1 |
Sample type |
RNA |
Source name |
HCC from FIV-transduced male mouse
Organism |
Mus musculus |
Characteristics |
strain: MF-1 outbred age: 433 days tissue: liver tumor gender: male fiv-transduced: yes mouse developed tumors: yes
Treatment protocol |
Liver tissue and tumors were flash frozen in liquid nitrogen and stored at -80 degrees until RNA extraction.
Growth protocol |
Pregnant mice (16 days gestation) were anaesthetized using isoflurane. Fetuses were removed from the womb and FIV-based viral particles were administered to the yolk sac vein of each fetus. The fetuses were then returned to the womb and were delivered naturally. Mice were kept under normal conditions until they were sacrificed.
Extracted molecule |
total RNA |
Extraction protocol |
Trizol extraction of total RNA was performed according to the manufacturer's instructions.
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).
Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Mouse Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.
Scan protocol |
GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G2500A.
Description |
M1-T2
Data processing |
Array data were normalized by the GCRMA algorithm using Affymetrix Expression Console. Data were transformed to log 2 scale and a threshold of 5 was applied.
