Sample GSM5061759 Query DataSets for GSM5061759
Status Public on Feb 04, 2021
Title rat-METH-METH challenge-1 rep
Sample type RNA

Source name total mRNA from PFC METH-METH group
Organism Rattus norvegicus
Characteristics strain: Sprague-Dawley
tissue: prefrontal cortex
gender: male
Treatment protocol The rats were administrated with METH (5 mg/kg, s.c.) or normal saline (NS) (1 ml/kg, s.c.) once daily in acute phase or for 7 days in development phase. On d8-14, the rats were housed in home cages, and nothing was injected in withdrawal phase. On d15, METH (1 mg/kg, s.c.) was used to challenge in the rats that were administrated with NS or METH in development phase, and then the locomotor activity was recorded for 60 min in challenge phase.
Extracted molecule total RNA
Extraction protocol The fluorescently amplified RNA targets were prepared from 2.5 μg of total RNA samples using the OneArray® Amino Allyl aRNA Amplification Kit (Phalanx Biotech Group)。
Label Cy5
Label protocol the amplified RNA argets were labeled with dye (Amersham Pharmacia, Piscataway, NJ, USA).

Hybridization protocol the labeled RNA targets were hybridized to the Rat Whole Genome OneArray® (ROA 1.1) (Phalanx Biotech Group) according to the Phalanx hybridization protocol.
Scan protocol The slides were scanned using an Axon 4000 scanner (Molecular Devices, Sunnyvale, CA, USA).
Data processing The Cy5 fluorescence intensity of each spot was analyzed using GenePix pro software (Molecular Devices).
The signal intensity of each spot was loaded into the Rosetta Resolver System® (Rosetta Biosoftware) for data analysis.
