Growth protocol |
For cultures, DO.11 T cells were isolated from donor spleens, activated with ova-pulsed splenocytes, skewed with cytokines and antibodies towards Th0, Th1 or Th2, and expanded with IL-2 for a total of 10 days (see also Mande et al JCI 2018 PMID: 29889098). For ex vivo analysis, DO.11 T cells were enriched from digested lesional skin tissue using Miltenyi CD3 bead isolation kits.
