Treatment protocol Eight week-old mice were fed a high-fat diet, (HFD; 58 kcal% fat w/sucrose, D12331; Research Diets, New Brunswick, NJ, USA) for 12 weeks.
Growth protocol All animals were housed at 22-24°C, with a 12:12-h light-dark cycle and ad libitum access to diet and water.
Extracted molecule total RNA
Extraction protocol Total RNAs were isolated using TRIzol (Invitrogen, MA, USA) according to the manufacturer’s instruction.
Label Cy3
Label protocol Cy3 labeled cRNA was prepared from 0.5 ug total RNA using the One-Color Low RNA Input Linear Amplification PLUS kit (Agilent) according to the manufacturer's instructions, followed by RNeasy column purification (QIAGEN, Valencia, CA).
 
Hybridization protocol Cy3 labeled cDNA was directly mix with hybridization buffer and Agilent blocking agent. Then, it was hybridized on microarrays for 40h at 65ºC in a rotating Agilent hybridization oven. After hybridization, microarrays were washed 1 minute at room temperature with GE Wash Buffer 1 (Agilent) and 1 minute with 37°C GE Wash buffer 2 (Agilent).
Scan protocol The array was scanned using a SureScan Microarray Scanner (Agilent).
Description Gene expression profiles of iWAT obtained from Senp2-aKO mouse
Data processing The intensity data of each individual hybridization were extracted and the quality was assessed with the Feature Extraction software (version 11.5.1) (Agilent). The log2-intensities of the probes from the arrays were normalized using the quantile normalization method.
